Carnitine: determination of total carnitine using a radioenzymatic assay.

چکیده

Acetylcarnitine and acylcarnitine are hydrolyzed by alkaline hydrolysis. The total carnitine is measured by incubation of the sample with 14CacetylCoA and carnitine acetyltransferase. The radioactive acetylcarnitine is separated from the unreacted radioactive acetylCoA using a Dowex 2 x 8 anion exchange column. The radioactive acetylcarnitine fraction is quantitated by counting in a liquid scintillation counter.

Plasma free and total carnitine measured in children by tandem mass spectrometry.

Free and total carnitine quantification is important as a complementary test for the diagnosis of unusual metabolic diseases, including fatty acid degradation disorders. The present study reports a new method for the quantification of free and total carnitine in dried plasma specimens by isotope dilution electrospray tandem mass spectrometry with sample derivatization. Carnitine is determined b...

Spectrophotometry of carnitine in biological fluids and tissue with a Cobas Bio centrifugal analyzer.

A spectrophotometric method for carnitine has been adapted to the Cobas Bio centrifugal analyzer. The addition of carnitine to a system containing carnitine acetyltransferase (EC 2.3.1.7) and acetyl-CoA gives rise to the formation of CoA. The system is coupled to 5,5'-dithiobis(2-nitrobenzoate) (DTNB). Assay response varied linearly with concentration of carnitine over a wide concentration rang...

An improved and simplified radioisotopic assay for the determination of free and esterified carnitine.

The radioisotopic assay for carnitine first described by Cederblad and Lindstedt (Clin. Chim. Acta. 37:235-543, 1972) and modified by Bohmer et al (Clin. Chim. Acta. 57:55-61, 1974) has been improved and simplified. As a result, the assay yields a linear response over a wide range of carnitine concentrations without the need for excessive amounts of labeled acetyl-CoA. In addition, it will meas...

Carnitine concentrations in term and preterm newborns at birth and during the first days of life.

Carnitine plays an important role in energetic metabolism. The aim of the study was to characterize the carnitine status in term and preterm newborns with respect to gestational age, birth weight, haematocrit and red blood cell count (RBC). The effect of nutrition on carnitine levels in the first week of life was also studied. Total blood pool of free carnitine (FC), acylcarnitines (AC) and tot...

Seminal carnitine and acetylcarnitine content and carnitine acetyltransferase activity in young Maremmano stallions.

The reproductive characteristics and seminal carnitine and acetylcarnitine content as well as carnitine acetyltransferase activity of young Maremmano stallions (n=25) are reported. The stallions were subjected to semen collection in November and January; in each trial two ejaculates were collected 1h apart. The total motile morphologically normal spermatozoa (TMMNS) and the progressively motile...
